The hospital was established by an Act of the legislature of Ontario in 1952 and was called the Ontario Cancer Institute. Initially, the hospital is located at 500 Sherburne Street, close to the former Wellesley Hospital.
In the 1990s, during a phase of structural redesign of healthcare services, PMH amalgamated with The Toronto Hospital, which itself was a combination of the Toronto General Hospital and the Toronto Western Hospital.
